Will the +1/+1 counters get rid of the -1/-1 counter or will Woodfall Primus have a -1/-1 counter and 2 +1/+1 counters

It's non creature destruction, not non land. Add a creature kill and you put them back to the begining. No enchantments, artifacts, lands, or planeswalkers. This combo leaves only creatures alive.

Eventually then, you destroy your own non-creature permanents including claws of gix, ashnod's altar and mighty emergence...
